Pathogenic for Von Hippel-Lindau syndrome; Chuvash polycythemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.11:g.(?_10183598)_(10206573_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the VHL protein in which other variant(s) (p.Arg64Pro) have been determined to be pathogenic (PMID: 9663592, 17102083, 17661816, 19336503, 24555745). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with VHL-related conditions. This variant results in the deletion of exons 2-3 and part of exon 1 (c.67_*14924delinsCCGAG) of the VHL gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product.
