Pathogenic for Curry-Hall syndrome; Ellis-van Creveld syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000004.11:g.(?_5564575)_(5758109_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 1-11 of the EVC gene, which includes the initiator codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in EVC are known to be pathogenic (PMID: 23220543). Isolated whole-gene deletions of EVC have not been reported in the literature. However, larger copy number events that include this gene have been reported (PMID: 18454448, 25174843). For these reasons, this variant has been classified as Pathogenic.
