Pathogenic for Renal carnitine transport defect — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.9:g.(?_131726438)_(131728974_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the SLC22A5 protein in which other variant(s) (p.Ala380Val) have been determined to be pathogenic (PMID: 23520115, 33757571). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with SLC22A5-related conditions. This variant results in the deletion of exon 8 and part of exon 7 (c.1109_1451-394del) of the SLC22A5 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in SLC22A5 are known to be pathogenic (PMID: 9916797).
