Pathogenic for Ichthyosis linearis circumflexa — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.9:g.(?_147204224)_(147516554_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the SPINK5 gene has been identified. Loss-of-function variants in SPINK5 are known to be pathogenic (PMID: 11511292, 11841556). The boundaries of this event are unknown as they extend beyond the assayed region for this gene and therefore may encompass additional genes. A similar copy number variant has been observed in individual(s) with Netherton syndrome and/or primary immunodeficiency disease (PMID: 16601670, 31795557, 34138484). For these reasons, this variant has been classified as Pathogenic.
