Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.11:g.(?_162394314)_(162475226_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 5-6 of the PRKN gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in PRKN are known to be pathogenic (PMID: 10072423, 20301651, 22956510). A similar copy number variant has been observed in individual(s) with autosomal recessive early-onset Parkinson's disease (PMID: 10824074, 21993715, 27182553, 28672806). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. For these reasons, this variant has been classified as Pathogenic.
