Pathogenic for LAMA2-related muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.11:g.(?_129465026)_(129465245_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the LAMA2 protein in which other variant(s) (p.Arg222Ser) have been determined to be pathogenic (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. A similar copy number variant has been observed in individual(s) with congenital muscular dystrophy (PMID: 24611677). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is a gross deletion of the genomic region encompassing exon(s) 5 of the LAMA2 gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame.
