Pathogenic for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma; Arrhythmogenic right ventricular dysplasia 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.11:g.(?_7583166)_(7587903_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of part of exon 24 (c.5671_*1792delinsAGAACAGTCTT) of the DSP gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. This variant has not been reported in the literature in individuals affected with DSP-related conditions. This variant disrupts a region of the DSP protein in which other variant(s) (p.Thr2634Lysfs*4, p.Gln2765Alafs*23, p.Thr2733Serfs*14, p.Lys2693Profs*3) have been determined to be pathogenic (PMID: 11063735, 21859740, 28527814). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
