Pathogenic for Duchenne muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.10:g.(?_30322696)_(33038337_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the DMD protein in which other variant(s) (deletion of exon 64) have been determined to be pathogenic (PMID: 22510846, 24504883, 24770780, 31705731). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. A similar copy number variant has been observed in individual(s) with Duchenne muscular dystrophy (PMID: 31705731). This variant is a gross deletion of the genomic region encompassing exon(s) 2-79 of the DMD gene, which includes the termination codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product.
