Uncertain significance for Joubert syndrome 15 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000007.13:g.(?_130038777)_(130039476_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with CEP41-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. ClinVar contains an entry for this variant (Variation ID: 848331). This variant results in the deletion of part of exon 11 (c.973+404_1077del) of the CEP41 gene. While this variant is not anticipated to result in nonsense mediated decay, it likely alters RNA splicing and results in a disrupted protein product.
