Pathogenic for Cystic fibrosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000007.13:g.(?_117144287)_(117144437_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 2 of the CFTR gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame. For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Ser50 amino acid residue in CFTR. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 10612827, 10875853, 25697318). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. A similar copy number variant has been observed in individuals with cystic fibrosis, pancreatic insufficiency, and congenital absence of vas deferens (PMID: 15520400, 16931591, 17448246, 22658665).
