Pathogenic for Fructose-biphosphatase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000009.11:g.(?_97355088)_(97365840_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of part of exon 7 (c.840_*10575del) of the FBP1 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. This variant has not been reported in the literature in individuals affected with FBP1-related conditions. This variant disrupts a region of the FBP1 protein in which other variant(s) (p.Glu281Lys) have been determined to be pathogenic (PMID: 23881342, 27101822, 29016355, 29774539). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
