Uncertain significance for Dilated cardiomyopathy 1W — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000010.10:g.(?_75862397)_(75863602_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of part of exon 15 (c.2023-1181_2047del) of the VCL gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in VCL cause disease. This variant has not been reported in the literature in individuals affected with VCL-related conditions. ClinVar contains an entry for this variant (Variation ID: 1784596). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
