NC_000011.9:g.(?_44129263)_(44745049_?)del was classified as Pathogenic by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Isolated whole-gene deletions of ALX4 have not been reported in the literature. However, larger copy number events that include this gene have been reported (PMID: 17290930, 24376213, 24780589). A gross deletion of the genomic region encompassing the full coding sequence of the ALX4 gene has been identified. Loss-of-function variants in ALX4 are known to be pathogenic (PMID: 11137991, 24668755, 25963140). The boundaries of this event are unknown as they extend beyond the assayed region for this gene and therefore may encompass additional genes.
