Pathogenic for Charcot-Marie-Tooth disease type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000011.9:g.(?_9878231)_(9902092_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 649478). This variant has been observed in individual(s) with Charcot-Marie-Tooth disease (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant results in the deletion of exon 18 and part of exon 19 (c.1929+15398_2137del) of the SBF2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in SBF2 are known to be pathogenic (PMID: 12687498, 25873783).
