It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with ATM-related conditions. This variant disrupts the substrate binding site of the ATM protein, which required for the DNA damage-induced association between ATM and its target proteins, including BRCA1,p53 (PMID: 15713674, 19781682). While functional studies have not been performed to directly test the effect of this variant on ATM protein function, this suggests that disruption of this region of the protein is causative of disease. Loss-of-function variants in ATM are known to be pathogenic (PMID: 23807571, 25614872). For these reasons, this variant has been classified as Pathogenic.
