NC_000012.11:g.(?_6554691)_(6560069_?)dup was classified as Pathogenic for Lymphoproliferative syndrome 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant results in a copy number gain of the genomic region encompassing part of exons 2 and 5 and all of exons 3-4 (c.238_547dup) of the CD27 gene. While the exact position of this variant cannot be determined from the data, sub-genic copy number gains are generally in tandem (PMID: 25640679). This variant is predicted to be out-of-frame, and may result in an absent or disrupted protein product. Loss-of-function variants in CD27 are known to be pathogenic (PMID: 25843314). This variant has not been reported in the literature in individuals affected with CD27-related conditions. A similar copy number gain has been confirmed to occur in-tandem in at least one individual (Invitae). This increases the likelihood that that this copy number gain also occurs in tandem and disrupts protein function. For these reasons, this variant has been classified as Pathogenic.
