Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_1401373)_(1412477_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a deletion of the genomic region encompassing 1-7 and part of exon 8 (c.-594_552del) of the GNPTG gene, and includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. This is expected to result in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with GNPTG-related conditions. Loss-of-function variants in GNPTG are known to be pathogenic (PMID: 19370764, 20301784). For these reasons, this variant has been classified as Pathogenic.
