Pathogenic for Duchenne muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.10:g.(?_32613975)_(32792683_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exons 8-12 and part of exon 13 (c.649+34927_1501del) of the DMD gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in DMD are known to be pathogenic (PMID: 16770791, 25007885). This variant has not been reported in the literature in individuals affected with DMD-related conditions. This variant disrupts a region of the DMD protein in which other variant(s) (p.Asp451Tyr) have been determined to be pathogenic (PMID: 22776072). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing.
