Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_56897677)_(56906303_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exons 1-6 and part of exon 7 (c.-1468_896del) of the SLC12A3 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SLC12A3 are known to be pathogenic (PMID: 20848653, 22009145, 25841442). A similar copy number variant has been observed in individual(s) with Gitelman syndrome (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 945564). For these reasons, this variant has been classified as Pathogenic.
