This variant disrupts a region of the TSC2 protein in which other variant(s) (p.Thr1068Ile) have been determined to be pathogenic (PMID: 21309039). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with TSC2-related conditions. This variant results in the deletion of exon 29 and part of exons 28 and 30 (c.3142_3531) of the TSC2 gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame.
