NC_000016.9:g.(?_68862109)_(68867341_?)del was classified as Pathogenic for Hereditary diffuse gastric adenocarcinoma by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the CDH1 protein in which other variant(s) (p.Glu836*) have been determined to be pathogenic (PMID: 29798843). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant has not been reported in the literature in individuals affected with CDH1-related conditions. This variant results in the deletion of exon 15 and part of exons 14 and 16 (c.2197_2588delinsGGCAAT, p.Arg733Glyfs*15) of the CDH1 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to disrupt the C-terminus of the protein.
