This deletion eliminates the WD40 functional domain, which is encoded by exons 6 to 13 of the PALB2 mRNA. The WD40 domain is required for normal PALB2 protein function (PMID: 16793542, 17200671,¬†19423707), and loss-of-function variants that disrupt this domain have been classified as pathogenic (Invitae). This variant has not been reported in the literature in individuals with PALB2-related conditions. This variant is a complex sequence change that deletes the genomic region encompassing exons 7-13 and part of exon 6¬†of the PALB2 gene¬†and inserts 240 nucleotides (c.2569_*9102delins240). It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product.
