Pathogenic for Fanconi anemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_89805009)_(89807294_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the FANCA protein in which other variant(s) (p.Arg1400His) have been determined to be pathogenic (PMID: 17924555, 29098742). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. A similar copy number variant has been observed in individual(s) with Fanconi anemia (PMID: 22178060). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is a gross deletion of the genomic region encompassing exon(s) 38-43 of the FANCA gene, which includes the termination codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product.
