Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_18054799)_(18058245_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exons 41-45 and part of exon 40 (c.7745_8224+176delinsCTCCCCGGAACGGCTGCTGGTAC) of the MYO15A gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in MYO15A are known to be pathogenic (PMID: 17546645). This variant has not been reported in the literature in individuals affected with MYO15A-related conditions. This variant disrupts a region of the MYO15A protein in which other variant(s) (p.Val2697) have been determined to be pathogenic (PMID: 23208854, 30622556, 32860223, 33398081).
