NC_000017.10:g.(?_33427972)_(33430307_?)del was classified as Pathogenic for Breast-ovarian cancer, familial, susceptibility to, 4 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the RAD51D protein in which other variant(s) (p.Arg300Aspfs*10) have been determined to be pathogenic (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with RAD51D-related conditions. This variant results in the deletion of exons 9 and 10 and part of exon 8 (c.704_987del) of the RAD51D gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product.
