Pathogenic for Leber congenital amaurosis 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_6337219)_(6337438_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the AIPL1 protein in which other variant(s) (p.Cys89Arg) have been determined to be pathogenic (PMID: 20702822, 23737531, 27268253, 28973376; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant is a gross deletion of the genomic region encompassing exon(s) 2 of the AIPL1 gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame. This variant has not been reported in the literature in individuals affected with AIPL1-related conditions.
