Loss-of-function variants in BRCA1 are known to be pathogenic (PMID: 20104584). This variant results in the deletion of exon 11 and part of exon 10 (c.4090_4185+236del) of the BRCA1 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. A similar deletion involving exon 11 and part of exon 10, known as exons 11 and 12 in the literature, have been observed in individuals with breast cancer (PMID: 18703817, Invitae).
