Pathogenic for Spongy degeneration of central nervous system — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_3397716)_(3402618_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exon 6 and part of exon 5 (c.707_*236del) of the ASPA gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. This variant has not been reported in the literature in individuals affected with ASPA-related conditions. This variant disrupts a region of the ASPA protein in which other variant(s) (p.His244Arg) have been determined to be pathogenic (PMID: 12638939, 16854607). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
