This variant disrupts a region of the TP53 protein in which other variant(s) (p.Arg337Cys) have been determined to be pathogenic (PMID: 9150393, 9704930, 9704931, 12826609, 17606709, 20128691, 21343334). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. A similar copy number variant has been observed in individual(s) with Li-Fraumeni syndrome (PMID: 7936651). It has also been observed to segregate with disease in related individuals. This variant is a gross deletion of the genomic region encompassing exon(s) 10 of the TP53 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to disrupt the C-terminus of the protein.
