Pathogenic for Multiple congenital anomalies-hypotonia-seizures syndrome 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000018.9:g.(?_59755969)_(59781892_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 15-25 of the PIGN gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in PIGN are known to be pathogenic (PMID: 24253414, 27038415). This variant has not been reported in the literature in individuals affected with PIGN-related conditions. For these reasons, this variant has been classified as Pathogenic.
