Pathogenic for Juvenile polyposis syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000018.9:g.(?_48556583)_(48593567_?)del, citing Invitae Variant Classification Sherloc (09022015): A similar copy number variant has been observed in individual(s) with juvenile polyposis syndrome (PMID: 18823382). This variant is a gross deletion of the genomic region encompassing exon(s) 1-10 of the SMAD4 gene, which includes the initiator codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SMAD4 are known to be pathogenic (PMID: 16152648, 16436638, 22810475). This variant is also known as promoter to coding exon 8. For these reasons, this variant has been classified as Pathogenic.
