Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015259.6(ICOSLG):c.657C>G (p.Asn219Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine, which is neutral and polar, with lysine, which is basic and polar, at codon 219 of the ICOSLG protein (p.Asn219Lys). This variant is present in population databases (rs778848090, gnomAD 0.005%). This missense change has been observed in individual(s) with combined immunodeficiency or refractory CMV viremia (PMID: 30498080, 34694545). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
