NM_004972.4(JAK2):c.1340T>A (p.Ile447Asn) was classified as Uncertain significance for Thrombocythemia 3 by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015. This variant lies in the JAK2 gene (transcript NM_004972.4) at coding-DNA position 1340, where T is replaced by A; at the protein level this means replaces isoleucine at residue 447 with asparagine — a missense variant. Submitter rationale: The missense variant c.1340T>A(p.Ile447Asn) in JAK2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The observed variant is absent in gnomAD exomes database. This variant has not been submitted to the ClinVar database. The amino acid change p.Ile447Asn in JAK2 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. Multiple lines of computational evidence (Polyphen- benign, SIFT- tolerated and MutationTasterpolymorphism) predicts a conflicting evidences on protein structure and function for this variant. The amino acid Ile at position 447 is changed to a Asn changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as Uncertain Significance (VUS).
