Uncertain significance for Duane-radial ray syndrome — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_020436.5(SALL4):c.635C>T (p.Pro212Leu), citing ACMG Guidelines, 2015: The observed missense c.635C>T(p.Pro212Leu) variant in SALL4 gene has not been reported previously as a pathogenic variant nor a benign variant, to our knowledge. The p.Pro212Leu variant has been reported with allele frequency of 0.002% in gnomAD Exomes. This variant has not been submitted to the ClinVar database. The amino acid change p.Pro212Leu in SALL4 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid Pro at position 212 is changed to a Leu changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as a Variant of Uncertain Significance (VUS).
