NM_002024.6(FMR1):c.838G>A (p.Glu280Lys) was classified as Uncertain significance for Abnormality of the nervous system; Fragile X syndrome by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015. This variant lies in the FMR1 gene (transcript NM_002024.6) at coding-DNA position 838, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 280 with lysine — a missense variant. Submitter rationale: The observed missense c.838G>A(p.Glu280Lys) variant in FMR1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in gnomAD Exomes. The amino acid Glu at position 280 is changed to a Lys changing protein sequence and it might alter its composition and physico-chemical properties. The amino acid change p.Glu280Lys in FMR1 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The variant is predicted as damaging by SIFT. For these reasons, this variant has been classified as Uncertain Significance.
