This variant lies in the POMT1 gene (transcript NM_001077365.2) at coding-DNA position 1680, where G is replaced by C; at the protein level this means replaces tryptophan at residue 560 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tryptophan, which is neutral and slightly polar, with cysteine, which is neutral and slightly polar, at codon 582 of the POMT1 protein (p.Trp582Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with POMT1-related conditions (PMID: 16575835). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 3242).
