Uncertain significance for Hereditary cancer-predisposing syndrome; Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005359.6(SMAD4):c.568G>A (p.Ala190Thr), citing Ambry Variant Classification Scheme 2023: The p.A190T variant (also known as c.568G>A), located in coding exon 4 of the SMAD4 gene, results from a G to A substitution at nucleotide position 568. The alanine at codon 190 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species; however, threonine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
