This variant lies in the LZTR1 gene (transcript NM_006767.4) at the canonical splice donor site of the intron immediately after coding-DNA position 791, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.791+2T>C intronic variant results from a T to C substitution two nucleotides after coding exon 8 in the LZTR1 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site. RNA studies have demonstrated that this alteration results in a transcript predicted to lead to a protein with an in-frame insertion of 31 amino acids; however, the exact functional impact of the inserted amino acids is unknown at this time (Ambry internal data).
