NM_006767.4(LZTR1):c.362A>C (p.His121Pro) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H121P variant (also known as c.362A>C), located in coding exon 4 of the LZTR1 gene, results from an A to C substitution at nucleotide position 362. The histidine at codon 121 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
