This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 118, where A is replaced by T; at the protein level this means converts the codon for lysine at residue 40 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The p.K40* pathogenic mutation (also known as c.118A>T), located in coding exon 2 of the NF1 gene, results from an A to T substitution at nucleotide position 118. This changes the amino acid from a lysine to a stop codon within coding exon 2. This alteration was observed in 1 of 77 Taiwanese/Chinese patients meeting diagnostic criteria for NF1 (Lee MJ et al. Hum Mutat, 2006 Aug;27:832). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
