NM_001042492.3(NF1):c.72A>T (p.Lys24Asn) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K24N variant (also known as c.72A>T), located in coding exon 2 of the NF1 gene, results from an A to T substitution at nucleotide position 72. The lysine at codon 24 is replaced by asparagine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
