NM_006009.4(TUBA1A):c.509C>G (p.Ser170Cys) was classified as Uncertain significance for Lissencephaly due to TUBA1A mutation by 3billion, citing ACMG Guidelines, 2015: The variant is not observed in the gnomAD v4.1.0 dataset. Predicted Consequence/Location: Missense changes are a common disease-causing mechanism. In silico tool predictions suggest damaging effect of the variant on gene or gene product [3Cnet: 0.99 (> 0.75, sensitivity 0.96 and precision 0.92)]. The same nucleotide change resulting in the same amino acid change has been previously reported to be associated with TUBA1A-related disorder (ClinVar ID: VCV003237530). However, the evidence of pathogenicity is insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
