Uncertain significance for Aldosterone-producing adenoma with seizures and neurological abnormalities — the classification assigned by Clinical Genomics Laboratory, Washington University in St. Louis to NM_001128840.3(CACNA1D):c.3275T>A (p.Met1092Lys), citing ACMG Guidelines, 2015: The CACNA1D c.3275T>A (p.Met1092Lys) variant, to our knowledge, has not been reported in the medical literature and is absent from the general population (gnomAD v.2.1.1), indicating it is not a common variant. This variant is predicted to occur in an alpha helix in a transmembrane domain (Pinggera A and Striessnig J. PMID: 26842699), changes a non-polar methionine to a positively charged lysine, and computational predictors indicate that the variant is damaging, evidence that correlates with impact to CACNA1D function. However, due to limited information, and based on ACMG/AMP guidelines for variant interpretation (Richards S et al., PMID: 25741868), the clinical significance of this variant is uncertain at this time.
