Uncertain significance for Beck-Fahrner syndrome; Abnormality of the musculoskeletal system — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_001287491.2(TET3):c.3301G>C (p.Val1101Leu), citing ACMG Guidelines, 2015: The missense variant c.3301G>C p.Val1101Leu in the TET3 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is novel not in any individuals in gnomAD Exomes and 1000 Genomes. The amino acid Valine at position 1101 is changed to a Leucine changing protein sequence and it might alter its composition and physico-chemical properties. The variant is predicted as damaging by SIFT. The amino acid change p.Val1101Leu in TET3 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
