NM_000352.6(ABCC8):c.1238C>G (p.Thr413Ser) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the ABCC8 gene (transcript NM_000352.6) at coding-DNA position 1238, where C is replaced by G; at the protein level this means replaces threonine at residue 413 with serine — a missense variant. Submitter rationale: Variant summary: ABCC8 c.1238C>G (p.Thr413Ser) results in a conservative amino acid change located in the ABC transporter type 1, transmembrane domain (IPR011527) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251476 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1238C>G has been reported in the literature in one individual affected with diabetes. The report does not provide unequivocal conclusions about association of the variant with Familial Hyperinsulinism. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 32027066). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
