NM_002225.5(IVD):c.389C>G (p.Ser130Cys) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: IVD c.389C>G (p.Ser130Cys) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 251460 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.389C>G has been reported in the literature as a mild-IVA variant with an ambiguous annotation, as c.398C>G without specifying the corresponding protein impact in at-least one reportedly asymptomatic homozygous newborn individual identified by newborn screening for Isovaleryl-CoA Dehydrogenase Deficiency (example, Mutze_2021). These report(s) do not provide unequivocal conclusions about association of the variant with Isovaleryl-CoA Dehydrogenase Deficiency. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 33496032). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
