Submitter rationale: Variant summary: FLNC c.2003_2007delinsGT (p.Asp668_Lys669delinsGly) results in an in-frame deletion-insertion that is predicted to delete one amino acid from the protein and also cause changes in one amino acid. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 5' splicing donor site. Four predict the variant creates a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 628588 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
