Submitter rationale: Variant summary: POLG c.1760_1765delinsTGGGCA (p.Pro587_Pro589delinsLeuGlyThr) results in an in-frame deletion-insertion that is predicted to cause changes in two amino acids within the encoded protein. It is part of a multinucleotide combination of 89868870-G-A (c. 1760C>T, p.Pro587Leu) and 89868865-G-T (c.1765C>A, p.Pro589Thr). The variant allele was found at a frequency of 8e-06 in 250060 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1760_1765delinsTGGGCA has been reported in the literature in an individuals affected with Mitochondrial DNA Depletion Syndrome - POLG Related (Uusimaa_POLG_Epilepsia_2013).
