Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000245.4(MET):c.124C>A (p.Gln42Lys), citing Ambry Variant Classification Scheme 2023: The p.Q42K variant (also known as c.124C>A), located in coding exon 1 of the MET gene, results from a C to A substitution at nucleotide position 124. The glutamine at codon 42 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
