Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001082486.2(ACD):c.1313T>A (p.Leu438His), citing Ambry Variant Classification Scheme 2023: The p.L524H variant (also known as c.1571T>A), located in coding exon 12 of the ACD gene, results from a T to A substitution at nucleotide position 1571. The leucine at codon 524 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
